Automated microscopy reveals complex biology across all length scales – from organelles to organisms

High content imaging (HCS) is the rapid, automated acquisition of microscopic fluorescence images in conjunction with automated image analysis software. Such experimental workflows are the standard in the drug discovery process of pharmaceutical companies, but are also highly applicable to academic research for visualization and rich quantitation of biological structures.

Technological advances in HCS allow researchers to follow complex, multidimensional processes at the cellular and intracellular level, both within cells and whole organisms. Multicolor fluorescence labeling allows for multiplexing to investigate biological processes and biochemical pathways. Dynamic cellular activities can readily be followed over prolonged culture time courses by taking advantage of the automated nature of HCS. Furthermore, novel 3D cell culture techniques, such as spheroids and organoids, and even whole organisms, such as zebrafish, are amenable to HCS. These studies make use of z-stacks to gather volumetric information as these multicellular structures respond to small molecule compounds.

In this seminar, we will highlight case studies of biological applications from current clients using the Hermes imaging system for:

· SPHEROIDS in multi cell-type co-culture to investigate metastatic cancer cell invasion

· MACROPHAGE immunological studies in response to yeast and HIV infection

· LABEL-FREE IMAGING of stem cells and other colony forming cells

· ORGANELLE and FOCAL ADHESION characterization at high resolution

· ZEBRAFISH in the context of infection and cell-type response to drug treatment

The Hermes WiScan® from IDEA Bio-Medical is the fastest multi-channel fluorescence HCS microscope and we have optimized it for speed, throughput and flexibility. It is coupled with WiSoft® Athena image analysis software constructed for biologists with the aim of enabling you to develop quantitative, image-based assays for phenotypic profiling. Our newest Hermes 24/7 integrated system comprises a robotic arm and a compact, dedicated cart for the creation of a fully autonomous experimental workflow from a single supplier. Together, they meet the requirements of modern biology for visualization of a multitude of cellular processes, while being easy to use with push-button operation.
